چکیده

METHODS: A total of 311 gram-positive cocci were tested, composed of 200 clinical isolates and 111 challenge strains. ID was performed on Staphylococci (171 strains), Enterococci (125 strains), and Streptococci (15 strains). AST was performed on all Staphylococal strains, and an Enterococcus faecalis (75 strains) and E. faecium (33 strains). The ID results were compared to API 20 Staph and API 20 Strep (bioMérieux, Lyon, France). Discrepant results were repeated in duplicate and remaining discrepant isolates were sent to the BDDS laboratory. For AST results the following drug classes (drugs) were evaluated in comparison to the NCCLS recommended standard broth micro-dilution method: Aminoglycosides (7), Beta-lactam antibiotics (6), Beta-lactam/beta-lactamase inhibitors (4), Carbapenems (2), Cephems (17), Folate antagonists (2), Quinolones (10), Glycopeptides (2), Macrolides / Lincosamides (7), Others (6). Discrepant results were repeated in duplicate, all data were evaluated as defined by the SM guidelines [1].

Evaluation of the Merlin MICRONAUT system for rapid direct susceptibility testing of gram-positive cocci and gram-negative bacilli from positive blood cultures.

Bloodstream infections are life-threatening conditions which require the timely initiation of appropriate antimicrobial therapy. We evaluated the automated Merlin MICRONAUT system for rapid direct microtiter broth antimicrobial susceptibility testing (AST) of gram-positive cocci and gram-negative bacilli from BACTEC 9240 bottles with positive blood cultures in comparison to the standard method ...

Evaluation of the LightCycler Staphylococcus M GRADE kits on positive blood cultures that contained gram-positive cocci in clusters.

We evaluated the Roche LightCycler Staphylococcus M(GRADE) kits to differentiate between Staphylococcus aureus and coagulase-negative staphylococci in blood cultures growing clusters of gram-positive cocci. Testing 100 bottles (36 containing S. aureus), the assay was 100% sensitive and 98.44% specific for S. aureus and 100% sensitive and specific for coagulase-negative staphylococci.

Rapid identification of obligately anaerobic gram-positive cocci using high-performance liquid chromatography.

High-performance liquid chromatography was evaluated as a rapid means of identifying obligately anaerobic gram-positive cocci of medical interest. Isolates were inoculated into a defined chemical medium consisting primarily of amino acids and were incubated aerobically for 1 h at 35 degrees C. After removal of organisms, the supernatant fluids were derivatized for 1 min at room temperature by t...

Achromopeptidase for lysis of anaerobic gram-positive cocci.

Achromopeptidase, which has potent bacteriolytic activity for most of the gram-positive aerobic bacteria, was for the first time used for the lysis of anaerobic cocci. Most of the lysozyme-resistant gram-positive anaerobic cocci were lysed with this new enzyme. Peptococcus magnus was the only organism tested resistant to achromopeptidase. P. saccharolyticus was quite unusual because it was very...

Vitek 2 automated identification system and Kocuria kristinae.

We have read with great interest the article by Ben-Ami et al. (2) about the misidentification of coagulase-negative staphylococci (CNoS) as Kocuria spp. by the Vitek 2 system (bioMérieux, Marcy l’Etoile, France). They warned that a clinical specimen growing Kocuria should raise suspicion of CNoS infection. In their study, they made use of the Vitek 2 ID GPC gram-positive identification card. I...
